Influence of hydrogen dioxide and chloramine B on biofilms of Koch’s bacillus

Biofilm formation is favorable for the survival of bacteria inhabiting aqueous environments. The study was designed for investigation of the effect of disinfectants, containing active substances – chlorine and hydrogen peroxide – on cholera vibrios of O1 serogroup in biofilm and planktonic forms. Eight Vibrio cholerae O1 strains of different origin and toxigenicity were taken into experiment. Disinfectants ("Chloramin B", hydrogen peroxide) were used in concentrations 0.1–0.5 % with 5–60 minute exposure at 23–25 °C. The biofilm formation was monitored on cover-slips or plastic plates. The viability of V. cholerae cells in the form of biofilm and in planktonic form was evaluated by growth (in CFU) on the Marten agar (pH 7.7) with incubation at 37 °C. Exopolysaccharide in biofilm composition was detected in vitro through successive staining by Congo red and fuchsine. Biofilms formed by test strains were visualized by microscopy. Adhesion of vibrios was revealed with biofilm formation on cover-slips and plates. After 5-day cultivation the biofilms demonstrated confluent growth on the Marten agar, V. cholerae planktonic cell concentration was 1105–1106 CFU/ml. It was also noted that V. cholerae O1 strains in the form of biofilm were resistant to 0.1–0.2 % hydrogen peroxide during 5–10 min in contrast to planktonic cells. In concentration 0.5–1.0 % and with 15 minute or more exposure hydrogen peroxide exhibited bactericidal activity on biofilm and planktonic cells. Treatment by 0.1–0.5 % "Chloramine B" for 5 min had no effect on vibrio biofilms. Unlike nontoxigenic strains, ctx+ strains in planktonic form showed no growth after 15-minute treatment. Thus, the biofilm forms of V. cholerae O1 demonstrated higher resistance to the tested disinfectant concentrations in comparison with planktonic form. Besides, toxigenic strains are more sensitive to disinfectants than nontoxigenic.

New poly-guanidines are innovating desinfectants with prolonged antimicrobic effect

In the present article there is shown findings of antibacterial, sanitary, engineering and technical aspects of polyalkylene- guanidines (PAG) as an innovating class of national prolonged-release antimicrobial agents. Summarized data concerning polymer influence of PAG and their structure to the complex of sanitary characteristics and to their antimicrobic effect. There are represented substances of microbiological study of several water-soluble salts of polyhexamethylated guanidine and polyether guanidines concerning gram-positive and gram-negative bacteria, mold fungi, microflora, which damage people’ skin covers. There were represented results of toxicity studies, which showed their hypotoxicity (4-th class of skin danger); there was showed relation between toxicological characterization from chemical structure of polymeric compound and also from residual value of monomers. There were showed examples of long-lasting effectiveness of desinfectants, which were used in medicine, food industry and for skin cover antimicrobic use.

The use of growth media of a new format for on-site control of disinfection quality

The report represents some results of a comparative study of sanitary-indicative test microorganisms detection efficacy in disinfection quality assessing in items under veterinary sanitary control using ready-made differential diagnostic growth media of a new format, namely Petrifilms, and some purpose-designed devices NovaStreak for rapid semi-quantitative detection and/or identification of potentially pathogenic or pathogenic microorganisms. The new format media are shown to have high detectability levels for sanitary-indicative test organisms with the detection efficacy for S. aureus in washings from absorbent and/or non-absorbent surfaces using 3M™ Petrifilm™ Staph Express Count Plate (STX) being not inferior to that one achieved when using the classical bacteriological method. Furthermore, application of the new format growth media does not require involvement of highly skilled professionals or using specially equipped laboratories, and allows on-site assessment of the efficiency of disinfection campaigns and some correction in case the obtained results are improper.

Safety estimation of impregnated organophosphorous compound tissue, which were dedicated to fight against clothes pediculosis

One of the most effective way to fight against clothes lices is use impregnated organophosphorous compound (FOS), underlinen. There was investigated percutaneous action of fenthion in dose of 20, 30 mpg and malathion in dose of 15, 50, 150, 300, 600 and 1200 mpg by two methods: by the way of emulsion substance applications to white rats’ and rabbits’ skin by the help of impregnated fabric body-clothes by agent. The exposure duration is 14–28 days. Time-exposure is 5 times per week for 4 houres a day. Biomarker of toxic action of FOS is activity of laborotary animals’ blood cholinesterase. By the way, there were measured activity of ALT, AST, AP, containig TP, albumins, urea, creatinine in blood serum. There were estimated the condition of nervous system of white rats. There was showed that boundaries of percutaneous action, which were estimated by different methods are 30 and 600 mpg for fenthion and malathion. There was proved high irritability of weanling rats to epicutaneous effect of malathion, comparing with nobilous rats. There were proved safety estimation of impregnated tissue including FOS, which is necessary for impregnation of underset.

There were represented statistics of morphology and biology of assassin and bed bugs, statistics about amplification reasons of bed bugs worldwide, statistics concerning development of multi-drug resistant bed bugs (C. lectularius) population in Russia and the ways against resistant populations.

Resistance investigation of german cockroaches (Blattella germanica L.) to insecticides from different classes by topic method

In laboratory environment there was assessment of resistance to insecticides from different chemical classes of Blattella germanica L. Race, derived from population, which were collected on food item Moscow-city (race RМ1). This race was under cultivation in laboratory environment during 4 generations without insecticides selection. There was approved that sensitive race of Blattella germanica L. of SNIID, cultivated in Institute and is sensitive to fipronil as international analog of sensitive races. There was determined that cockroaches of race RM1 are high-resistant to pyrethroids (50–140x), to phenylpyrazoles (9–12x) and tolerant or resistant to organophosphorous compounds (5–15x), carbamates (4x), avermectins (3–10x), also are sensitive to neonicotinoides (0,7–1x). There was determined that cockroaches’ multiple resistance of race RM1 to insecticides form 5-th classes of chemicals.

Pediculicidal activity of pyrethroids and organophosphorous compounds during tissue impregnation

lothes’ impregnation by insecticides is the most effective preventive measure, which protects population boom of clothes louses in organized associations. There are recommended emulsion concentrates on the basis of organophosphorous compound (FOS), also mixtures of fenthion with permethrin in different proportions, because of lices’ resistance development to permethrin for impregnation of tissues. There were estimated beginning of palsy state’ rate, insects’ death rate, remedies’ effective concentration depending on their formula and water capacity of worked tissue. Findings are the basis criteria of efficiency’ estimation of impregnated tissue by insecticides: violent action – 100 % death of adult insects and larvas of III instars during 60 min. of exposure dose on impregnated tissue (cotton sheeting) after 24 h.; time period of motion activity stopping of all zooids in experiment – not more than 180 min. during contact with impregnated tissue.
