Disinfectants as reagents for DNA-decontamination

One of the problems facing the application of amplification-based techniques is false-positivity due to DNA-contamination. The aim of this study was to determine the usefulness of different types of disinfectants for DNA-decontamination. Laboratory experiments were designed to evaluate of quaternary ammonium compounds (QACs), combinations of QACs with aldehydes and QACs with alcohol, amphotensides and chlorine disinfectants for their ability to destroy or irreversibly modify the DNA, preventing its subsequent amplification. Also methods of purification of DNA from disinfected samples were compared. It was found that amplicons exposed to QACs and amphotensides couldn’t be detected by electrophoresis. However, samples of amplicons with these disinfectants gave positive PCR results after DNA extraction, indicating the preservation of intact DNA, and thus QACs and amphotensides cannot be applied for elimination of contaminating nucleic acids. This study showed that only chlorine disinfectants at a concentration of at least 0,2 percent of available chlorine can be used for the DNA-decontamination.
Key words: PCR, DNA-contamination, DNA-decontamination, disinfectants.

Sanitizers are on the basis of aldehyds (analytical survey)

In survey there was represented comparative analysis of aldehyds as the most reactive class of organic compounds, which were used in healthcare organizations. There were summarized updated data concerning aldehyds’ structures, mechanism of biocidal activity, toxic level, action on workpieces, materials, including medical devices. There are defined benefits of disinfectants on the basis of aldehyds: their match with different kinds of materials, low corrosivity, biodegradability in environment. It was demonstrated that in spite of some lacks such as expressed irritation action, aggressive odor, sensitize and cumulative effects, fixation of organic impurities on workpieces, the necessity of preliminary objects’ clear and by the help of wide range of antimicrobic action – medications on the basis of aldehyds (mainly on glutaric and ortho-phthalic) are useful for disinfection and sterilization.
Key words: disinfection, sterilization, sanitizers, aldehyds, antimicrobic action, toxic level.

Methods of analysis of aldehydes (analytical review)

Aldehydes are widely used in various fields of science and technology. Sometimes they are used as reagents in organic synthesis, but they are most widely used as substances obtained for chemical disinfection and sterilization. Glutaraldehyde is widely used for these purposes, later the using of ortho-phthalaldehyde has been started. Glyoxal and other aldehydes are used much less frequently due to their lower antimicrobial activity. Analysis aldehydes extremely difficult because of high reactivity of these compounds, moreover, some deteriorate during storage. For a quantitative analysis of aldehydes using various methods – by titration until HPLC-MS. In this paper all the basic techniques that have been proposed for quantitative determination of aldehydes in various objects, including air.
Key words: glutaraldehyde, glyoxal, ortho-phthalaldehyde, methods of analysis.

Food scent as the safety method against flies in the buildings

Taking into account high degree of epidemiological importance of flies, which are agents’ vector of different diseases, that is why the search of effective measures and methods of their population decrease. As far as preventive measures don’t guarantee 100 % result therefor it’s necessary to implement destructive measures such as to use imagocides, in different cases – larvicides, but there are good reasons to use them together. Type of preparative form is selected by object category, for example, for some there can be used irrigation method, aerosol, for others (babe’s and food’s) are recommended to use scents, sticky gel compositions and so on, including the safest preparative forms. The most important it’s for hospitals and different health stations, also for using in passenger transportations objects: railway, marine, air, provided by the presence of people.
Key words: synanthropic flies, food scents, infections, azamethiphos, cifenitrin, metomil, fipronil, acetamiprid, thiamethoxam, imidacloprid.
